dc.description.abstract | With the intensification of energy crisis and environmental problems, biomass energy has become the focus of energy strategy of various countries because of its advantages of sustainability and renewable energy, microbial fuel cells have been widely concerned because of its development prospects in sewage treatment and clean electricity production. Microbial Fuel cells (MFC) can degrade pollutants while producing electrical energy through the metabolic action of electrogenic microorganisms. Shewanella spp is the most widely studied electrogenic microorganism in the laboratory, which is mainly allogeneic metal-reducing bacteria. For example, the microbe can generate an electric current, with the help of an Extracellular Electron Transfer (EET) mechanism. In this study, it was believed that TBSR3 gene of Shewanella is crucial for the formation of outer membrane pigment protein. Therefore, the function of TBSR3 gene of Shewanella was analyzed by using in-frame deletion mutation method to knock out the TBSR3 gene of Shewanella, and the changes in electrical production capacity of defective mutant strains were accurately detected, so as to verify the viewpoint. Colony PCR and agarose gel electrophoresis were also used to verify the experiments, and the colonies were screened with the help of nutrient deficiency types and resistance genes. | uk |
